Is it possible to ship to a pickup point with Cash On Delivery?

Yes - unfortunately, Shopify does not allow adding a surcharge for COD to the shipping cost. If you charge a COD fee, simply include it in the shipping price.

COD for courier only

By default, Shopify also does not allow making payment methods conditional on the selected shipping method. If you want to offer COD only for courier delivery (and prepayment for pickup points), use a payment method hiding app from the Shopify App Store and hide COD when the selected shipping method is a pickup point.

Our app does not modify payment methods - it only adds pickup point shipping methods.
